While outcomes for children with T-cell acute lymphoblastic leukemia have improved dramatically, survival rates for patients with relapsed/refractory disease remain dismal. Prior studies indicate that glucocorticoid resistance is more common than resistance to other chemotherapies at relapse. In addition, failure to clear peripheral blasts during a prednisone prophase correlates with an elevated risk of relapse in newly diagnosed patients. Here we show that intrinsic GC resistance is present at diagnosis in early thymic precursor T-ALLs as well as in a subset (...) of non-ETP T-ALLs. GC-resistant non-ETP T-ALLs are characterized by strong induction of JAK/STAT signaling in response to interleukin-7 stimulation. Removing IL7 or inhibiting JAK/STAT signaling sensitizes these T-ALLs, and a subset of ETP T-ALLs, to GCs. The combination of the GC dexamethasone and the JAK1/2 inhibitor ruxolitinib altered the balance between pro- and anti-apoptotic factors in samples with IL7-dependent GC resistance, but not in samples with IL7-independent GC resistance. Together, these data suggest that the addition of ruxolitinib or other inhibitors of IL7 receptor/JAK/STAT signaling may enhance the efficacy of GCs in a biologically defined subset of T-ALL.Leukemia advance online publication, 30 May 2017; doi:10.1038/leu.2017.136. In this paper, gathering several topics present in the work of Newton da Costa, we propose a rigorous foundation for a possible formulation of scientific theories according to the semantic approach. Following da Costa, as a first step we develop a general theory of structures; inside this theory we show how we can characterize formal languages as particular kinds of structures, more specifically, as free algebras. Next we discuss how we can link a language to a structure, with which we (...) can formulate the axioms that are intended to capture the theory of the structure. Nesse artigo eu proponho, partindo da distinção entre conceitos semânticos e conceitos ontológicos, a separação entre descrição semântica e postulação de entidades. A postulação de uma entidade implica o fornecimento de um critério de identificação, isto é, a verdade de uma asserção de identidade entre dois dispositivos de designação. Isso parece indicar que a existência da entidade seria garantida pela simples existência de um critério de identificação. Porém, esta é uma condição de significatividade para um termo referencial: que ele codifique (...) ou esteja associado a uma descrição identificadora. Tal descrição identificadora não pode, de modo algum, ser confundida com uma condição de existência para a entidade em questão. (shrink)

Veremos, nos dois autores, a crítica implacável das pretensões racionalistas e a denúncia dos impasses e das frustrações resultantes das escolhas da modernidade.

Contractualism is one of the most promising ‘centers of gravity’ in business ethics. In this guest editorial we provide a concise roadmap to the field, sketching contractualism’s historic and disciplinary antecedents, the basic argumentative structure of the contract model, and its boundary conditions. We also sketch two main dimensions along which contributions to the contractualist tradition can be positioned. The first dimension entails positive versus normative theorizing – does a given contribution analyze the world as it is or how it (...) ought to be? The second dimension involves four different levels of analysis that are commonly employed in contractualist business ethics: the nano, micro, meso, and macro levels. Resumo: O presente artigo tem como objetivo analisar três aspectos do tema do amor na obra de Friedrich Nietzsche. Para tanto, parte de dois pressupostos: [1] o sentimento do amor deve ser entendido como parte do projeto de crítica à metafísica e aos idealismos presentes na moral, na religião e na arte; e como consequência, [2] como crítica às noções do amor que remete à tradição socrático-platônico, judaico-cristã e moderna. Para tanto, os três aspectos daí derivados, mostram que o amor (...) em Nietzsche 1] não é falta, [2] nem negação de si, [3] nem está baseado na ideia de posse; muito pelo contrário, [1] amor é exuberância, [2] depende da fruição de si e [3] está baseado nas diferenças próprias e desigualdades naturais entre os indivíduos. Tal procedimento analítico chega à afirmação da amizade como amor superior, cujo conteúdo remente a um sentimento de homens nobres, precisamente aquilo que se poderia chamar, em Nietzsche, de um grande amor.: This article aims to analyze three aspects of the theme of love in Friedrich Nietzsche’s work. For that, it starts from two assumptions: [1] the feeling of love must be understood as part of the project of criticism of metaphysics and the idealisms present in morals, religion and art; and as a consequence, [2] as a critique of the notions of love that refer to the Socratic-Platonic, Judeo-Christian and modern tradition. The three aspects derived from it, show that love in Nietzsche [1] is not lack, [2] nor self-denial, [3] nor is it based on the idea of possession; quite the contrary, the opposite link, [1] love is exuberance, [2] depends on the enjoyment of oneself and [3] is based on self-differences and natural inequalities between individuals. Such an analytical procedure comes to the affirmation of friendship as a superior love, whose content refers to a feeling of noble men, precisely what could be called, in Nietzsche, great love. Background: We like to think about sexual activity as something fixed, basic and primal. However, this does not seem to fully capture reality. Even when we relish sex, we may be capable of mentalizing, talking, voluntarily postponing orgasm, and much more. This might indicate that the central control mechanisms of sexual activity are quite flexible and susceptible to learning mechanisms, and that cortical brain areas play a critical part. Objective: This study aimed to identify those cortical areas and mechanisms most (...) consistently implicated in sexual activity. Design: A comprehensive review of the human functional neuroimaging literature on sexual activity, i.e. genital stimulation and orgasm, is made. Results: Genital stimulation recruits the classical somatosensory matrix, but also areas far beyond that. The posterior insula may be particularly important for processing input from the engorged penis and coordinating penile responses. Extrastriate visual cortex tracks sexual arousal and responds to genital stimulation even when subjects have their eyes closed. The ventromedial prefrontal cortex is also tightly coupled to sexual arousal, but low activity in this area predicts high sexual arousal. Conclusion: This review has indicated cortical sites where activity is moderated by tactile genital inflow and high sexual arousal. Behavioral implications are discussed and where possible the relevance for learning mechanisms is indicated. Overall, it is clear that the cerebral cortex has something to say about sexual activity. Life and contributions of eminent philosophers from Andhra Pradesh, India.

In the climate of concern about high medical costs, the relationship between the trade and professional aspects of medical practice is receiving close scrutiny. In the United Kingdom there is talk of increasing privatisation of health services, and in the United States the Federal Trade Commission (FTC) has attempted to define medicine as a trade for the purposes of commercial regulation. The Supreme Court recently upheld the FTC charge that the American Medical Association (AMA) has been in restraint of trade (...) because of ethical strictures against advertising. The concept of profession, as it has been analyzed in sociological, legal, philosophical, and historical perspectives, reveals the importance of an ethic of service as well as technical expertise as defining characteristics of professions. It is suggested that the medical profession should pay more attention to its service ideal at this time when doctors are widely perceived to be technically preoccupied. (shrink)

People with disabilities face inequalities in mental wellbeing, for which social exclusion is a contributing factor. Musical activities offer a promising but complex intervention, making impacts on a population with highly varied characteristics and needs challenging to capture. This paper reports on a mixed methods, person-centered study investigating a community music intervention for such a population. Three groups of adult service users with varied disabilities (either physical, learning, or both), took part in weekly music workshops in different locations. Music staff, (...) housing and resource center staff, as well as participants and members of their families, took part in semi-structured interviews. A quantitative measure administered by service staff was used to rate service users’ social development. Two lay researchers, both individuals with a disability contributed to all aspects of the study. Interviews were analyzed through thematic analysis. Improvements in individuals’ self-expression, confidence, mood, and social skills were consistent with previous findings. Differences in effect between centers included: Group 1, some of whom had previous experience of workshops, showed an improvement in musical skills; Group 2 showed a mixed response, some participated with enthusiasm but others chose art activities over music workshops; Group 3 had lasting positive impact, this group had very limited opportunities for music due to their rural location. Quantitative analysis showed significant increase over all groups in communication, interaction with others, and joint attention. The intervention was beneficial for participants in separate locations in similar ways, but also highlighted that context and prior experience mediated effects in distinct ways. The lay researchers enhanced the qualitative analysis by emphasizing (1) the importance of recognizing participants’ self-expression in non-verbal modes of communication and (2) the importance of having music staff with a disability to provide a positive role model. This paper proposes that mixed methods person-centered research is the most suitable approach to capture and understand the multiple and varied effects of this complex intervention for a diverse group of participants. In the second edition Preface of Religion Within the Bounds of Bare Reason Kant responds to an anonymous review of the first edition. We present the first English translation of this obscure book review. Following our translation, we summarize the reviewer's main points and evaluate the adequacy of Kant's replies to five criticisms, including two replies that Kant provides in footnotes added in the second edition. A key issue is the reviewer's claim that Religion adopts an implied standpoint, described using (...) transcendental terminology. Kant could have avoided much confusion surrounding Religion, had he taken this review more seriously. Abstract In this paper, we explore how the application of technological tools has reshaped food production systems in ways that foster large-scale outbreaks of foodborne illness. Outbreaks of foodborne illness have received increasing attention in recent years, resulting in a growing awareness of the negative impacts associated with industrial food production. These trends indicate a need to examine systemic causes of outbreaks and how they are being addressed. In this paper, we analyze outbreaks linked to ground beef and salad greens. (...) These case studies are informed by personal interviews, site visits, and an extensive review of government documents and peer-reviewed literature. To explore these cases, we draw from actor-network theory and political economy to analyze the relationships between technological tools, the design of industrial production systems, and the emergence and spread of pathogenic bacteria. We also examine if current responses to outbreaks represent reflexive change. Lastly, we use the myth of Prometheus to discuss ethical issues regarding the use of technology in food production. Our findings indicate that current tools and systems were designed with a narrow focus on economic efficiency, while overlooking relationships with pathogenic bacteria and negative social impacts. In addition, we find that current responses to outbreaks do not represent reflexive change and a continued reliance on technological fixes to systemic problems may result in greater problems in the future. We argue that much can be learned from the myth of Prometheus. In particular, justice and reverence need to play a more significant role in guiding production decisions.
